CUSABIO's product CSB-E12121Fh is a ready-to-use ELISA kit suitable for the measurement of growth hormone (GH) in fish serum, plasma, or tissue homogenates. It can detect fish GH in the range of 312.5 pg/mL-5000 pg/mL. And it has been validated in multiple parameters, including sensitivity, specificity, precision, linearity, and recovery.

The mechanism of this assay is based on the competitive inhibition enzyme immunoassay technique. In this assay, the kit has been pre-coated with an antibody specific to GH. Samples and standards are respectively pipetted into wells with the Biotin-conjugated GH. GH in the sample or in the standards competes with the biotin-labeled GH for binding to the pre-coated GH antibody. After washing, HRP-Avidin conjugate is added to wells. The solution turns blue after the addition of the TMB substrate. The color reaction is terminated and the solution turns from blue to yellow when adding the stop solution. The color intensity is negatively correlated to the amount of GH bound in the initial step and is measure at 450 nm via a microplate reader.

Growth hormone (GH) is a protein hormone synthesized and secreted by the anterior lobe of the pituitary gland. It regulates somatic growth by stimulating the production of the IGF1 in the liver and other organs. It also participates in the modulation of metabolism and entry homeostasis.

Materials provided
  • A 96-well Assay plate --The 96-well plate has been pre-coated with anti-Fish GH antibody.
  • Standard (6 x 1 ml) --Dilute the standard at dilution series, read the OD values, and then draw a standard curve.
  • HRP-avidin (1 x 6 ml) --Catalyze the TMB to elicit a chromogenic reaction.
  • Biotin-conjugated GH (1 x 6 ml) --Bind to the precoated antibody and act as the detection antigen.
  • Sample Diluent (2 x 20 ml) --Reconstitute the standard and dilute the sample to an appropriate concentration.
  • Wash Buffer (20x concentrate) (1 x 15 ml) --Wash away unbound or free substances.
