This Human N-terminal pro-brain natriuretic peptide (NT-proBNP) ELISA Kit is used to quantitatively determine mouse NT-proBNP concentrations in serum, plasma, or tissue homogenates.NT-proBNP is a cardiac hormone secreted by atria and ventricles and released into circulation upon ventricular stretch or ischemia. Congestive heart failure (CHF) is the chief culprit to elevate the levels of NT-proBNP. NT-proBNP is a very good marker of cardiac function and a strong prognostic parameter in patients with coronary heart disease and heart failure. Lucian MURESAN etc. demonstrated that NT-proBNP levels could become a useful ventricular arrhythmia marker for assessing the arrhythmic risk in patients with systemic sclerosis. This kit employs the quantitative sandwich enzyme immunoassay technique to measure the levels of human NT-proBNP in the sample. Antibody specific for NT-proBNP has been pre-coated onto the microplate. Standards and samples are pipetted into the wells and any NT-proBNP present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated NT-proBNP antibody is added to the wells. After washing, avidin conjugated HRP is added to the wells, forming an antibody-antigen-enzyme-labeled antibody complex. Following a wash to remove any unbound HRP-avidin, the TMB substrate solution is added to the wells, and color develops into blue. The color changes from blue to yellow after the addition of stop solution into the wells. The intensity of the color is in proportion to the amount of NT-proBNP bound in the initial step. The absorbance (OD value) was determined by the microplate reader at the wavelength of 450nm, and the concentration of mouse NT-proBNP was calculated through the standard curve. Each manufactured lot of this ELISA kit is quality tested for criteria such as sensitivity, specificity, precision, linearity, and lot-to-lot consistency.
