The Human Osteopontin (OPN) ELISA Kit quantities OPN levels in human serum, plasma, urine, or tissue homogenates. OPN is a secreted matricellular cytokine synthesized by cells involved in bone morphogenesis. It is highly up-regulated in an acute and chronic inflammatory environment. And it is implicated in physiological and pathophysiologic processes, including biomineralization and bone remodeling, cellular immunity and wound healing, and tumorigenesis. OPN has been demonstrated to promote proliferation, angiogenesis, and metastasis of tumor cells. It also signals through integrin and CD44 receptors. OPN interacts with CD44 via an RGD-independent manner to express different functional activities in chemotaxis of macrophages, cell adhesion of fibroblasts, and cytoprotection and regulation of T helper type 1 cells and bone marrow cells.
This kit employs the sandwich-ELISA mechanism in conjugation with OPN antibody-OPN antigen-specific binding as well as HRP-TMB chromogenic reaction to measure the concentrations of OPN in the samples. The kit is characterized by high sensitivity, strong specificity, good linearity, recovery with 81%-106%, precision less than 10%. Refer to the product instructions for more validation information.

- A micro ELISA plate ---The 96-well plate has been pre-coated with an anti-human OPN antibody. This dismountable microplate can be divided into 12 x 8 strip plates.
- Two vials lyophilized standard ---Dilute a bottle of the standard at dilution series, read the OD values, and then draw a standard curve.
- One vial Biotin-labeled OPN antibody (100 x concentrate) (120 μl/bottle) ---Act as the detection antibody.
- One vial HRP-avidin (100 x concentrate) (120 μl/bottle) ---Bind to the detection antibody and react with the TMB substrate to make the solution chromogenic.
- One vial Biotin-antibody Diluent (15 ml/bottle) ---Dilute the Biotin-antibody.
